The human gene was inserted upstream of the translation start site and downstream of the promoter of the mouse host gene. The loxP site flanked neomycin resistance gene that was also inserted, was deleted through cre-mediated recombination. This leaves an allele with the Diphtheria Toxin sensitive human gene under control of the mouse promoter, rendering the cells where this allele is expressed sensitive to DT-induced cell death.
