A cDNA encoding rat TNNC1 (troponin C, cardiac/slow skeletal) carrying an L48Q (CTG->CAG) mutation and a FLAG epitope tag was placed under the regulatory control of the mouse Myh6 (myosin, heavy polypeptide 6, cardiac muscle, alpha) promoter and the tetracycline responsive element (tetO). Translation is terminated by a human growth hormone polyadenylation signal. When active, the transgene elicits ~40% incorporation of the mutant cardiac troponin C into the cardiac muscle sarcomere.
