Exon 4 was targeted with the insertion of a COIN (conditional by inversion) cassette. The cassette consists of a splice acceptor site and a LacZ gene with polyA site flanked by lox71 and lox66 sites in head-to-head orientation. The cassette is inserted in inverted orientation compared to the target gene. Upon cre-mediated recombination the modified loxP sites will mediate irreversible inversion of the cassette and thus create a gene for a fusion between the 5' end of the target gene and the lacZ gene. Also inserted into exon 4, downstream of the COIN cassette but still in exon 4, was an FRT site flanked neomuycin resistance gene cassette and a splice acceptor site. This allele is conditional-ready and splits exon 4 into exon 4a and 4b which has no effect on the mature transcript. (J:240991)
