An FRT site flanked neomycin resistance gene cassette and a loxP site were inserted into intron 4. A second loxP site was inserted into intron 8. The neo cassette was removed through subsquent flp-mediated recombination. The targeted exons were deleted through cre-mediated recombination. This creates a null allele with exons 5a through 8, encompassing all the exons encoding the catalytic domain, deleted. The inclusion of exon 5a in the deletion means that this allele affects both the short splice variant, which ends with exon 5a, and the long variant. (J:240314)
