The transgene contains a mouse promoter Thy1.2 (Thyb) driving neuronal expression of a FRET-based ATP indicator (J:240165) composed of a variant of cyan fluorescent protein (mseCFP) linked with circularly permuted mVenus having the 173rd amino acid as its N terminus (cp173-mVenus) to either the N- or C terminus of the epsilon subunit of the Bacillus subtilis FoF1-ATP synthase Adenosine 5-Triphosphate (atpc). Four hydrophobic amino acid residues (Val-9, Leu-42, Phe-67, and Leu-78) of the epsilon subunit were replaced by hydrophilic residues to retard incorporation into the FoF1 and reduce nonspecific hydrophobic interaction with other proteins. In addition, M60Y, G131E, K132M and A133K mutations were introduced. This sensor is known as ATeam (adenosine 5-Triphosphate indicator based on Epsilon subunit for Analytical Measurements) 1.03YEMK (ATeam1.03YEMK) and can be used for measuring intracellular ATP levels in the millimolar range.
