A tandem duplication of the gene resulted from integration of a vector containing a neomycin selection cassette, a loxP site, the 5' end of the Hprt gene, and a homology region including all three exons of the targeted gene. The two copies of the gene are separated in the mouse chromosome by the vector sequence. Quantitative real-time PCR of total RNA extracts of liver and brain demonstrated ~2-fold greater transcript levels, relative to those of wild-type mice, in both tissues of mice with three gene copies and ~3-fold increase in mice with four copies.
