The transgenic construct contains a murine Myh6 gene promoter directing expression of an a modified dsRed1 cDNA fused to the mitochondrial targeting sequence from human COX8A followed by a human growth hormone polyadenylation signal. Transgene expression is detected predominantly in the heart ventricles, with lower expression in the atria, which is expected due to the lower number of mitochondria in the atrial cardiomyocytes. The modified DsRed1-E5 (or Timer) fluorescent protein contains two amino acid substitutions, V105A and S197T, which increase fluorescence activity and allow a color shift from green to red over 48 hours.
