The transgene consists of a construct that expresses amino acids 1751-2290 of the mouse Notch1 intracellular domain (NICDOP) with an N-terminal FLAG sequence and a modified estrogen receptor (ER) fused in frame at the C terminus. Expression of the construct is driven by a keratin 14 promoter cassette. Founder lines 5199.15A,B+C (CA3, low copy number) and 5199.21A,B+C (CA4, contains 21 copies of the transgene at high copy number) were established; both lines had a similar phenotype, the severity correlating with copy number.
