A loxP site upstream of exon 5, followed by a FRT-flanked puromycin resistance cassette downstream of exon 5 and another loxP site were inserted via homologous recombination. The puromycin resistance cassette was removed via Flp-mediated recombination leaving exon 5 flanked by loxP sites. (J:238323)
