Through homologous recombination a loxP site was inserted into intron 6 and an FRT site flanked neomycin resistance gene cassette and a second loxP site into intron 7. Subsequent cre-mediated recombination deleted out-of-frame exon 7 (which encodes part of the kinase domain), thus creating a null allele with premature stop codons in exon 8. PCR confirmed the absence of exon 7 in transcripts from this allele. (J:238171)
