A BAC (RP23-479M11) containing the Prop1 gene was modified by inserting a cassette composed of the sequence encoding a mammalian codon usage optimized cre recombinase (icre), the bovine GH polyadenylation signal and an Frt-flanked kanamycin selection marker, into the exon 2 in frame. Flpe excision removed the kanamycin selection marker. The modified BAC was injected into pronuclei of fertilized oocytes to generate transgenic mice. Three lines, 432, 506, and 517 were genereted, and all 3 lines exhibit cre activity throughout the pituitary anterior lobe at 3 weeks of age. Line 517 was used for studies because it best reflected endogenous Prop1 expression. (J:233898)
