An FRT-loxP-neomycin-FRT-loxP cassette is inserted upstream of exon 21 (encoding amino acids 929-985, the fifth transmembrane domain and pore). A third loxP is inserted downstream of exon 21. Flp-mediated recombination removed the FRT-flanked neo cassette leaving exon 21 floxed. (J:237834)
