A targeting vector containing a human HBEGF diphtheria-toxin receptor (DTR) cDNA linked in frame to the dsRed Express2 fluorescent protein by a 2A peptide, a FRT-flanked PGKneo cassette, a bovine growth hormone polyadenylation signal and two loxP sites, are used to disrupt the translation initiation codon in exon 2. DsRed fluorescence is detected in small intestine Paneth cells and colonic deep crypt secretory cells, and mimics the endogenous Reg4 gene expression pattern. No red fluorescence is detected in pancreas, stomach, or liver. (J:235576)
