Exon 5 was flanked by loxP sites. An FRT-flanked neomycin selection cassette was removed by breeding heterozygous floxed animals with actin-Flp recombinase mice, and the loxP-flanked exon 5 sequences were removed by breeding with zona pellucida promoter-driven Cre mice which express Cre in developing oocytes. Immunoblot analysis using anti-CP110 antibody on protein extracts confirmed the absence of protein expression in E14.5 homozygous mutant embryos. (J:232855)
