A mouse bacterial artificial chromosome (BAC) containing the entire Npas1 gene, was modified by inserting iCre-2A (cleavable fusion peptide sequence)-tdTomato and polyadenylation sequence from bovine growth hormone downstream of the ATG translation initiation codon, and removing Npas1 coding sequence. A transgenic insert made from this modified BAC was microinjected into the pronucleus of C57BL/6 fertilized eggs. The resulting Npas1-tdTom transgenic mice from founder line 1 were subsequently maintained on the C57BL/6J background. (J:237688)
