The targeting vector is designed to insert a loxP-and FRT-flanked neomycin resistance (neo) cassette upstream of exon 2 of the gene. A third loxP site was inserted downstream of exon 2. Flp-mediated recombination removed the FRT-flanked neo cassette leaving exon 2 floxed. (J:237493)
