Pronuclear-stage embryos were targeted with a zinc finger nuclease pair designed to target exon 4. A founder mouse was identified carrying a 2-bp deletion at the targeted site (GAACTGCTGTatGGACGAGC -> GAACTGCTGTGGACGAGC), creating a reading-frame shift. Absence of the protein was confirmed by immunoblot analysis. (J:249809)
