The insertion of Velocigene cassette ZEN-Ub1 created a deletion of size 3091bp between positions 10180726-10183816 of Chromosome 3 (Genome Build37). Exons 1-4 were replaced by a lacZ sequence (in frame after the original ATG) and a loxP-neo-loxP cassette. Cre-mediated recombination removed the neo cassette. RT-PCR and Western blot analyses confirmed the absence of mRNA and protein expression in sciatic nerve endoneurium of P56 homozygous mutant mice. Coimmunostaining of Pmp2 and Mbp on P10 sciatic nerve cross sections confirmed the loss of Pmp2 expression in all Schwann cells with no detectable changes in Mbp expression. (J:136110, J:211344)
