A 124 base pair (bp) loxP site has replaced a PvuII restriction enzyme recognition site 76 bp past the translation termination codon, and a Pgk-neomycin phosphotransferase cassette flanked by frt sites and followed by a second loxP site has replaced an AccI recognition site 446 bp downstream of the stop codon. The loxP sites flank both the frt-flanked selection cassette and an ultraconserved sequence (UCS) in the 3' untranslated region (3'-UTR) of the mouse gene that mediates post-transcriptional repression of protein synthesis.
