The locus was targeted with a construct containing the ubiquitous CAG promoter, a floxed stop cassette and the human cDNA. The targeting construct also contains the promotor and exons of the human ortholog of the target locus, thus restoring the target locus. The stop cassette was removed through cre-mediated recombination, allowing the transcription of the inserted human cDNA. (J:233357)
