The microRNA sequence was replaced with a floxed neomycin resistance cassette. Cre-mediated recombination removed the selection cassette. The deletion results in a nonsense mutation in exon 3 and a premature stop codon on the opposite strand in Rtl1. Western blot analysis confirmed the absence of Rtl1 protein expression in conceptuses.
