A floxed neomycin resistance cassette was inserted in front of exon 3 and an additional loxP site after exon 6. Mice were mated with Ella-Cre Deleter mice to remove the neomycin resistance cassette as well as exons 3 to 6, leaving behind a single loxP site between exon 2 and 7. Western blot analysis confirmed absence of protein expression in brain lysates from adult homozygous mutant mice. (J:228428)
