A targeting vector was designed to insert a loxP upstream of exon 4 (containing the start codon), and an FRT-flanked neomycin resistance (neo) cassette, followed by a second loxP site, downstream of exon 5 gene. Cre-mediated recombination removed exons 4 through 5 and the neo cassette. Western blot and immunohistochemistry of prostrate and ovary confirmed that no protein was expressed in mutant mice.
