Sequence encoding a promoterless eGFP minigene, a loxP site and the PGKneo cassette is inserted upstream of exon 1 (containing the translation start site), and another loxP site followed by the PGK promoter is inserted downstream of exon 2. The entire targeted region is flanked by FRT sites. Recombination using Cre recombinase will remove the exons while allowing EGFP expression or FLP-mediated recombination will remove both the exons and the EGFP. (J:233743)
