A PGK-neomycin expression cassette has replaced 1.5 kbp of the 5' non-coding region and the first 3 exons of the gene. RT-PCR analysis detects no gene transcripts and western blot analysis confirms the absence of protein expression in pulmonary tissue of homozygous mutant mice; quantitative ELISA of 2-fold diluted serum from mice detects no protein in their circulation, and immunohistochemical analysis demonstrates absence of the protein in their arterial tissue.
