The transgene contains a single copy of the mouse tyrosinase gene, including all five exons and the 5' and 3' untranslated regions (UTRs), but lacks the locus control region (LCR) regulatory elements. Transgenic mice were generated by pronuclear injection of mouse oocytes with a 100-kb yeast artificial chromosome (YAC), YRT4. YRT4 was produced by deletion via homologous recombination of 150 kb of DNA, including the LCR and upstream sequence, from the 250-kb YAC YRT2, which was derived from a YAC library generated from strain C3H genomic DNA. Expression of the transgene is demonstrated by its (weak) partial rescue of the pigment phenotype of albino NMRI mice. (J:223068)
