The transgene contains a single copy of the entire mouse tyrosinase expression domain, including all five exons, the 5' and 3' untranslated regions (UTRs), and ~155 kb of 5' upstream sequences encompassing the locus control region (LCR) regulatory elements at -12 kb. Transgenic mice were generated by pronuclear injection of mouse oocytes with a 250-kb yeast artificial chromosome (YAC), YRT2, from a YAC library generated from strain C3H genomic DNA. Expression of the transgene is demonstrated by its complete rescue of the pigment phenotype of albino NMRI mice. (J:4498)
