Two amino acid substitutions (D121R;R122D) were made to change the highly conserved DRY motif to RDY to diminish G-protein signalling, and an internal ribosome entry site followed by a constitutively active Gnas coding sequence (due to a Q227L sequence change) was inserted just downstream of the endogenous coding sequence to give bicistronic expression.
