The 12-kb transgene, derived from the pPrHG "half genomic" vector, comprises the promoter and first exon of the mouse prion protein gene (Prnp); mouse exon 2 joined to exon 3, containing the entire prion protein open reading frame, from kudu (an antelope of the genus Tragelaphus); and the 3'-untranslated region (UTR) of the mouse gene. The encoded protein contains six copies of the amino-terminal octapeptide repeat sequence (6OR), rather than the usual five copies. Mice bearing this transgene express kudu 6OR prion protein from the mouse Prnp promoter in brain and other tissues. (J:32214, J:82809, J:232471)
