A gene trap containing an FRT flanked neomycin resistance cassette and tdTomato reporter, followed by loxP flanked exon 2 was inserted into intron 1 via homologous recombination. FLP-mediated recombination removed the FRT flanked neomycin cassette and tdTomato reporter, leaving exon 2 flanked by loxP sites. (J:225909)
