The HTGRS1006_A_4_H03 targeting vector was obtained from The International Knockout Mouse Consortium by The Jackson Laboratory. The L1L2_Pgk_P cassette was inserted at position 13615101 of Chromosome 8 upstream of the critical exon 3 (Build GRCm38). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by neomycin resistance gene under the control of the PGK promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of the targeted exon 3 at position 13614300. The critical exon 3 is thus flanked by loxP sites. Flp-mediated recombination removed the reporter cassette and left exon 3 floxed. (J:221899)
