A loxP site and F3-flanked puromycin resistance cassette were inserted upstream of the microRNA sequence. A second loxP site and an FRT-flanked neomycin resistance cassette were inserted downstream of the microRNA sequence. Flp-mediated recombination removed the selection cassette and left the microRNA sequence floxed. (J:221990)
