A loxP site, a copy of the human AU/U rich elements of the human FOS gene, mouse Edn1 3' UTR, two copies of transcriptional insulators from the chicken ACTB gene, neo cassette, loxP site and Adm2 3' UTR were inserted just downstream of the stop codon via homologous recombination. In the absence of cre expression, the presence of the AU/U rich element destabilizes the mRNA, reducing expression. Following cre mediated recombination the Adm2 3' UTR stabilizes the mRNA, increasing expression. (J:220962)
