Mice with this transgene express wild-type human hepatic lipase (LIPC) under control of the regulatory elements of the human apolipoprotein E (APOE) locus. The construct contains 3 kb of 5'-flanking sequence, exon 1, intron 1, and the first 6 nucleotides of exon 2 from the human APOE gene followed by the cDNA encoding human LIPC and, from the human APOE locus, the untranslated region of exon 4, 0.1 kb of 3'-flanking sequence, and the first hepatic control region. Western blot analysis using a rabbit polyclonal anti-human LIPC antiserum confirmed the presence of immunoreactive human hepatic lipase in sera of mice bearing this transgene. Analysis of postheparin plasma demonstrated hepatic lipase activity in the human physiologic range in transgenic mice also homozygous for the null Lipctm1Unc mutation. (J:229079, J:229080)
