A loxP site, splice acceptor site, wild-type mouse Syk cDNA, FRT flanked neo cassette, loxP site, splice acceptor site, and exon 2 with an R41A mutation were inserted via homologous recombination. In the absence of cre mediated recombination the wild-type gene is expressed. Following cre mediated recombination a mutant protein lacking binding to phosphotyrosine at the N-terminal SH2 is expressed. (J:220779)
