A floxed pgk-neo-STOP cassette, Loxl2 cDNA, IRES and EGFP were inserted via homologous recombination. Cre mediated recombination removed the pgk-neo-STOP cassette. Fluorescence imaging, semi-quantitative PCR, and Western blot analysis of whole-cell extracts and conditioned medium confirmed targeted overexpression in newborn homozygous mutant mice and derived MEFs.
