TALEN mediated recombination inserted a tdTomato reporter and triple polyadenylation cassette into exon 1. Detection of tdTomato fluorescence was specific to skeletal muscle and was not detected in other tissues, such as tendon. Real-time PCR analysis using primer pairs specific to the downstream exons 2 and 3 confirmed the absence of transcript in skeletal muscle as well as in liver. (J:217579)
