Lox2372 and loxP sites were inserted in intron 1 and an antisense oriented conditional inversion (COIN) module, a second set of lox2372 and loxP sites, and an FRT flanked neo cassette were inserted in intron 3 The two lox sites are in reverse complementation orientation, thus cre-mediated recombination will irreversibly invert the intervening sequence. The COIN module consists of a splice acceptor site and a T2A-GFP-pA cassette. Flp-mediated recombination removed the drug selection cassette. (J:217084)
