The transgene, constructed in the pBROAD3 vector (InvivoGen), encodes the 560 C-terminal amino acids of the mouse GAK (cyclin G associated kinase) protein, which comprise its clathrin binding domain and its HSPA8- (also called HSC70-) interacting J domain. Expression of the partial Gak cDNA is driven by a modified Gt(ROSA)26Sor promoter whose 5' untranslated region (5' UTR) is interrupted by an engineered intron; the expressed sequence is followed by the human beta-globin 3' UTR and polyadenylation signal. Immunoblot analysis using polyclonal rabbit antibodies to GAK demonstrated expression of the truncated protein in all tissues examined: brain, heart, kidney, liver and lung. (J:225348)
