R3A and K5A mutations were inserted in exon 1 and a floxed neo cassette was inserted into intron 1 via homologous recombination. The mutation in exon 1 disrupts interaction of the protein with subunits of the nucleosome remodeling and histone deacetylase complex. Cre mediated recombination removed the neo cassette. Western blot analysis of E13.5 heart lysates revealed comparable protein levels in wild-type, heterozygous, and homozygous mutant hearts. (J:216295)
