The construct contains the chicken beta-actin promoter (with cytomegalovirus immediate early enhancer) driving expression of the mitochondrial targeting sequence from Cox8a (cytochrome c oxidase subunit VIIIa) fused to a pH-sensitive fluorescent protein, Keima, flanked by attB sites. This construct was microinjected, along with the PhiC31 integrase mRNA, into the pronuclei of zygotes collected from Igs2tm3.1Luo mice. The Igs2tm3.1Luo allele contains three tandem attP sites. The PhiC31 integrase catalyzes recombination between the attB and attP sites. (J:101977)
