The targeting vector contains a 2A sequence, followed by a destabilized nuclear CFP (nuclear localized CFP fused with "PEST" sequence), 2A sequence, followed by destabilized TVA (TVA fused with "PEST" sequence), bGH polyA. An FRT-flanked PGK-NEO cassette was inserted adjacent to the STOP codon in exon 4, of the Fos gene. "PEST" sequence targets the protein for degradation by fusing amino acids 422-462 from the degradation domain of mouse ornithine decarboxylase to the C-terminal of the reporter/protein (in this case CFP and TVA).
