The targeting construct was designed (by site-directed mutagenesis) to substitute a serine for an alanine at codon 112 in exon 2. A loxP-STOP-loxP (LSL) cassette was inserted between exons 1 and 2. Cre-mediated recombination removed the LSL cassette allowing conditional expression of the S112A mutation. (J:223215)
