LoxP-flanked neomycin resistance cassettes were introduced both 5' and 3' of the exon that encodes the kinase active site (the fifth coding exon). The 5' neomycin cassette was excised with Cre recombinase during vector construction in E. coli. Cre-mediated recombination removed the 3' floxed neo cassette, leaving the kinase active site exon flanked by loxP sites. (J:101977)
