LoxP sites are inserted (1) within the first exon 5' of the initiator methionine codon, and (2) 3' of an SV40 polyadenylation signal that is inserted immediately 3' of the 3' UTR. Inserted downstream of the 3' loxP site are the coding region for CFP with a 3' HA tag, an FRT site, a phosphoglycerate kinase (PGK)-Neo cassette, a third loxP site and an FRT site. (J:264733)
