A Cys521Ser substitution was introduced into exon 18 by changing a TGT codon to a TCC codon and an F3 flanked puro selection cassette was introduced into intron 16 via homologous recombination. Flp mediated recombination removed the selection cassette. This mutation prevents hydrolysis of the enzyme's epoxyeicosatrienoic acid (EET) substrates. (J:211353)
