The mouse bacterial artificial chromosome (BAC) RP24-306A6 containing the entire Pvalb gene, was modified first by a BAC trimming method to remove the Ift27 gene coding region. The second BAC recombineering round inserted the tdTomato-WPRE-BGHpA (bovine growth hormone polyadenylation signal)-FRT-NEO-FRT vector directly downstream of the Pvalb initiation start codon. The FRT site flanked NEO was then removed by transient FLP recombinase expression. Correctly targeted BAC DNA was used as the source for the transgene, which was linearized to remove it from the vector. (J:101977, J:232250)
