By sequential targeting an IRES-EGFP and FRT flanked neo cassette were inserted downstream of the open reading frame. Flp-mediated recombination removed the neo cassette. Then a loxP site and neo cassette and loxP site were inserted around the N1 enhancer element. Cre-mediated recombination removed the N1 enhancer and neo cassette. Homozygous mice show loss of Sox2 expression in the zone at the caudal end of neural plate but expression is seen in the neural tube. (J:217059)
