The 180kb mouse bacterial artificial chromosome (BAC) ID#508P6, containing the entire Peg3 (or Pw1) gene, was modified by inserting an IRESnLacZ cassette, a SV40 polyadenylation sequence signal, and a FRT-flanked kanamycin selection cassette 5' of exon 9. The kanamycin cassette was excised before microinjection. LacZ reporter activity, mRNA expression and protein are coexpressed in the developing limbs, heart, somites, splanchinic mesoderm and floor and roof plates of the neural tube. (J:174391)
