An IRES-GFP-FRT-PGK-neo-FRT-loxP cassette was inserted immediately downstream of the stop codon in exon 12, along with a loxP site in intron 10. The PGK-neo cassette was removed via Flp-mediated recombination, leaving exons 11-12 and the IRES-GFP floxed. Expression of GFP was confirmed by flow cytometry and live cell imaging in megakaryocyte progenitor cells, megakaryoctyes, and LSK cells. (J:208120)
